Coronin 1A (CORO1A) is a WD-repeat, actin-binding protein predominantly expressed in leukocytes, where it is essential for actin cytoskeletal remodeling and cell migration. It forms a beta-propeller structure that interacts with F-actin, influencing Rac1 signaling, T-cell receptor-induced calcium flux, and thymic egress. CORO1A is critical for the survival and homeostasis of naive T cells and also contributes to phagosome dynamics in macrophages. Clinically, loss-of-function mutations in CORO1A are a well-established cause of autosomal recessive T-cell immunodeficiency, characterized by a severe reduction in peripheral T cells and increased susceptibility to viral and opportunistic infections.
