COMM domain containing 8 (COMMD8) is a member of the COMM domain-containing protein family, characterized by a conserved C-terminal domain that facilitates protein-protein interactions and nuclear transport. It functions as a critical adaptor protein, forming a stable heterodimeric complex with COMMD3 to prevent proteasomal degradation. This complex selectively recruits G protein-coupled receptor kinase 6 (GRK6) to chemoattractant receptors such as CXCR4, thereby promoting β-arrestin-mediated MAPK signaling and lymphocyte chemotaxis. Additionally, COMMD8 interacts with CCDC22 and Cullin1-RING E3 ubiquitin ligases to enhance the ubiquitination and turnover of IκB, leading to the activation of NF-κB-mediated transcription. Clinically, COMMD8 is significantly upregulated in non-small cell lung cancer, where it acts oncogenically to drive cell proliferation, migration, and resistance to apoptosis. Its expression levels in these contexts correlate with poor patient prognosis, making it a potential biomarker and therapeutic target in oncology.
