Baculoviral IAP repeat containing 3 (BIRC3), also known as cIAP2, is a multifunctional protein that belongs to the inhibitor of apoptosis (IAP) family and plays a crucial role in regulating apoptosis, inflammation, and immune signaling. Structurally, BIRC3 contains three baculoviral IAP repeat (BIR) domains, a ubiquitin-associated (UBA) domain, a caspase recruitment domain (CARD), and a RING finger domain, which collectively enable it to inhibit caspases and modulate various signaling pathways, including those involved in mitogenic kinase activity and inflammatory responses. BIRC3 interacts with tumor necrosis factor receptor-associated factors (TRAF1 and TRAF2), thereby inhibiting apoptosis and influencing cell survival, particularly under conditions such as serum deprivation. The protein is widely expressed across tissues, including immune and endocrine organs, and its function can be further diversified by alternative 3' untranslated regions (3' UTRs) that facilitate the assembly of specific protein complexes, impacting processes like B cell migration. Dysregulation or mutation of BIRC3 has been implicated in various diseases, notably certain leukemias and lymphomas, where altered BIRC3 function can contribute to abnormal cell survival and resistance to apoptosis.
