Centrosomal protein 57 like 1 (CEP57L1) is a centrosomal scaffold protein that localizes to the microtubule-organizing center and is essential for maintaining centrosomal architecture. Together with its paralog CEP57, it redundantly regulates centriole engagement during interphase, preventing precocious disengagement and abnormal centriole reduplication. CEP57L1 also plays a role in mitotic spindle organization by tethering the minus ends of spindle microtubules and facilitating their attachment to kinetochores. Clinically, reduced expression of CEP57L1 has been specifically associated with the congenital absence of the anterior and posterior cruciate ligaments in the knee, indicating a specialized role in musculoskeletal development.
