Cyclin dependent kinase 16 (CDK16) is a serine/threonine protein kinase of the cdc2/CDKx subfamily that regulates vesicle-mediated transport, exocytosis, and secretory cargo trafficking. Activated by cyclin Y family proteins, CDK16 is highly expressed in the brain and testis, where it is essential for normal spermatogenesis, neuronal differentiation, and dendrite development. It modulates physiological processes such as glucose-responsive insulin secretion and growth hormone release. Clinically, CDK16 is associated with X-linked hereditary spastic paraplegia and male infertility characterized by oligospermia. Furthermore, it is frequently overexpressed in various cancers, where it promotes cell proliferation, radioresistance, and the establishment of an immunosuppressive tumor microenvironment.
