CD33 molecule (CD33) is a type I transmembrane sialic-acid-binding immunoglobulin-like lectin primarily expressed on myeloid lineage cells, including monocytes, macrophages, and granulocyte precursors. Upon binding to sialylated ligands, CD33 recruits SHP-1 and SHP-2 phosphatases via its cytoplasmic ITIM motifs, delivering inhibitory signals that dampen cytokine production and maintain immune cells in a resting state. Pathologically, CD33 is highly expressed on the blasts of approximately 90% of patients with acute myeloid leukemia (AML), making it a primary target for immunotherapies such as gemtuzumab ozogamicin. Additionally, CD33 variants are associated with altered microglial function and increased risk in Alzheimer’s disease.
