TNF receptor superfamily member 12A (TNFRSF12A), also known as the TWEAK receptor (TWEAKR) or fibroblast growth factor-inducible immediate-early response protein 14 (FN14), is a protein encoded by the TNFRSF12A gene in humans. It is a unique member of the Tumor Necrosis Factor Receptor superfamily, characterized by its small size and highly regulated gene expression in both live organisms and in vitro settings. TNFRSF12A is a type I transmembrane protein with a cysteine-rich domain (CRD) and no death domain, yet it can induce a weak death cell signal. It is expressed in various human tissues, including the heart, placenta, lung, skeletal muscle, kidney, and pancreas, and plays a crucial role in promoting angiogenesis and the proliferation of endothelial cells. Increased expressions or interactions of TNFRSF12A have been correlated with several diseases, including acute ischemic stroke, Rheumatoid Arthritis, Systemic Lymphocytic Erythematosus (SLE), Multiple Sclerosis, and Cancer.
