CD160 molecule (CD160) is a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein expressed on NK cells, cytotoxic CD8+ T cells, and intestinal intraepithelial lymphocytes. It contains a single Ig-like V-type domain and binds broadly to classical and non-classical MHC class I molecules, as well as the TNF receptor family member HVEM. CD160 functions as a dual-purpose immune modulator, delivering co-inhibitory signals to CD4+ T cells while providing activating signals to NK cells via the PI3K-AKT-mTOR pathway to enhance cytokine production and cytotoxicity. Clinically, CD160 acts as an immune checkpoint; its dysregulation is linked to impaired immune function in cancer and HIV infection. It is currently being investigated as a therapeutic target for cancer immunotherapy and the management of pathological neoangiogenesis.
