Membrane metalloendopeptidase (MME), also known as neprilysin, CD10, or CALLA, is a zinc-dependent metalloprotease that cleaves peptides at the amino side of hydrophobic residues. It is a 100 kDa type II integral membrane protein with a highly conserved zinc-binding motif in its extracellular C-terminal domain. MME inactivates several peptide hormones, including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. It also degrades amyloid beta peptide, implicated in Alzheimer's disease. MME is expressed in various tissues, particularly abundant in the kidney, and serves as an important cell surface marker in diagnosing acute lymphocytic leukemia (ALL). Diseases associated with MME include Charcot-Marie-Tooth Disease, Axonal, Type 2T and Spinocerebellar Ataxia 43. Recent studies have shown that MME expression is positively correlated with systemic lupus erythematosus and may inhibit breast cancer occurrence.
