C-C motif chemokine ligand 17 (CCL17), also known as thymus and activation-regulated chemokine, is a secreted CC chemokine that plays a pivotal role in shaping type 2-biased immune responses. It is primarily produced by dendritic cells and other antigen-presenting cells to selectively attract CCR4- and CCR8-expressing CD4+ Th2 lymphocytes. CCL17 is essential for T-cell development in the thymus and the trafficking of mature T cells to sites of inflammation. Clinically, elevated CCL17 levels are a hallmark of Hodgkin lymphoma and various skin malignancies, where it facilitates the recruitment of Th2-rich inflammatory infiltrates. Additionally, increased circulating CCL17 is associated with cardiac hypertrophy and fibrosis, suggesting its potential as a therapeutic target in heart failure.
