Carbonic Anhydrase 4 (CA4) is a glycosylphosphatidylinositol (GPI)-anchored membrane glycoprotein encoded by the CA4 gene, part of the carbonic anhydrase family that catalyzes the reversible hydration of carbon dioxide to bicarbonate and protons, essential for pH regulation, ion transport, and CO₂ metabolism. Predominantly expressed in the kidney, retina, and capillary endothelium, CA4 facilitates bicarbonate reabsorption in renal tubules and maintains acid-base balance, while also contributing to retinal function, possibly via fluid homeostasis. Mutations in CA4 are linked to autosomal recessive retinitis pigmentosa (RP17), a progressive retinal degeneration causing blindness, and renal tubular acidosis, a disorder marked by impaired urinary acidification leading to systemic acidosis.
