Chromosome 9 open reading frame 78 (C9orf78) is a human protein involved in the late-stage regulation of pre-mRNA splicing. It functions by interacting with the spliceosome remodeling enzyme SNRNP200 (BRR2), binding to its coiled-coil domain to moderately inhibit helicase activity on U4/U6 di-snRNA. This interaction facilitates spliceosome maturation and influences 3'-splice site selection. C9orf78 also binds to U5 snRNA and displaces WBP4 from the SNRNP200 complex, ensuring proper alternative splicing patterns. Beyond its role in the spliceosome, C9orf78 interacts with FAM50A to upregulate the expression of asparagine synthetase (ASNS). This pathway is clinically significant in oncology, as the FAM50A-C9orf78 axis has been shown to enhance the migration, invasion, and brain metastasis of breast cancer cells. Consequently, C9orf78 is recognized as a key factor in promoting aggressive tumor phenotypes and metastatic progression.
