Transcription factor Jun (JUN), also known as c-Jun, is a key component of the activator protein 1 (AP-1) transcription factor complex, which regulates various cellular processes including proliferation, differentiation, and apoptosis. JUN is encoded by the JUN proto-oncogene and functions as a transcriptional regulator by forming homo- or heterodimers with other proteins like c-Fos. It is activated through phosphorylation by several kinases, including JNK (c-Jun N-terminal kinase), which enhances its transcriptional activity. Dysregulation of JUN has been implicated in numerous diseases, particularly cancer. Overexpression of c-Jun is frequently observed in aggressive tumors, such as non-small cell lung cancer and breast cancer, where it promotes survival and proliferation of malignant cells. Furthermore, aberrant JUN signaling contributes to inflammatory diseases and neurodegenerative disorders.
