Bone marrow stromal cell antigen 2 (BST2), also known as tetherin or CD317, is a type II transmembrane protein encoded by the BST2 gene located on chromosome 19p13.2. BST2 is constitutively expressed in mature B cells, plasma cells, and plasmacytoid dendritic cells, and is induced by interferon in other cell types. It functions as an antiviral host restriction factor by tethering enveloped viruses to the cell membrane, preventing their release and promoting their degradation. BST2 also plays a role in cell membrane integrity, lipid raft formation, and signaling pathways such as GRB2/ERK/BIM/Cas3 and PI3K/AKT, which are implicated in tumorigenesis, invasion, and metastasis. High BST2 expression is associated with poor prognosis in various cancers, including breast cancer, and is linked to increased risk of metastasis and resistance to anoikis. BST2 is also involved in immune regulation and has been implicated in diseases such as rheumatoid arthritis and viral infections.
