B-cell lymphoma 6 protein (BCL6) is a transcriptional repressor that belongs to the BTB/POZ (Broad-complex, Tramtrack, and Bric-à-brac/Pox virus and Zinc finger) family of proteins. It contains an N-terminal BTB/POZ domain, a central transcriptional repressor domain, and C-terminal zinc finger motifs that enable DNA binding. BCL6 is a master regulator of humoral immunity, playing a crucial role in the initiation and maintenance of germinal centers (GCs) during the immune response. It promotes the survival, proliferation, and differentiation of GC B cells by repressing genes involved in cell cycle arrest, apoptosis, and terminal differentiation. Dysregulation of BCL6 has been strongly implicated in the pathogenesis of B-cell lymphomas, particularly diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma (FL). Overexpression of BCL6 in these cancers contributes to uncontrolled proliferation, genomic instability, and immune evasion.
