Autophagy related 7 (ATG7) is an E1-like ubiquitin-like modifier-activating enzyme essential for macroautophagy and cytoplasm-to-vacuole transport. It activates ATG12 and ATG8/LC3 family proteins to drive ATG12-ATG5 conjugation and LC3 lipidation, which are critical steps for autophagosome membrane formation and cargo sequestration. Beyond its core role in autophagy, ATG7 modulates p53-dependent cell cycle pathways and participates in mitophagy, hematopoietic stem cell maintenance, and circadian control of metabolism. Pathogenic variants in ATG7 are established causes of autosomal recessive spinocerebellar ataxia 31, a neurodegenerative disorder characterized by Purkinje cell axonal dystrophy and impaired axonal autophagy.
