Angiopoietin 1 (ANGPT1) is a secreted glycoprotein that serves as a constitutive agonist for the endothelial tyrosine kinase receptor TIE2 (TEK). It is characterized by a C-terminal fibrinogen-like domain that mediates receptor binding and clustering, which in turn promotes endothelial cell survival, migration, and the stabilization of cell-cell junctions. ANGPT1 is essential for embryonic vascular development, vessel branching, and the recruitment of pericytes to ensure the formation of mature, non-leaky vasculature. Clinically, an imbalance in the ANGPT1/ANGPT2 ratio is implicated in pathological conditions such as sepsis-associated vascular leak, edema, and abnormal tumor angiogenesis. Enforced expression of ANGPT1 has been shown to normalize tumor vessels and strengthen the vascular barrier, highlighting its therapeutic potential.
