Aldehyde dehydrogenase 1 family member A1 (ALDH1A1) is a cytosolic enzyme primarily responsible for oxidizing a broad range of endogenous and exogenous aldehydes, including retinaldehyde to retinoic acid, which is essential for retinoid signaling and cellular differentiation, as well as detoxifying reactive aldehydes generated by lipid peroxidation and alcohol metabolism. ALDH1A1 is highly expressed in the liver and certain neuronal populations, where it also plays a key role in dopamine metabolism by converting toxic intermediates to less harmful products, thereby protecting neurons from oxidative stress. Clinically, ALDH1A1 is recognized as a robust marker for both normal and cancer stem cells, conferring properties such as self-renewal, chemoresistance, and tumor initiation, and its overexpression is linked to poor prognosis and therapy resistance in several cancers, including ovarian and lung cancer. Furthermore, ALDH1A1 deficiency or dysfunction is implicated in neurodegenerative diseases like Parkinson's disease due to impaired detoxification of neurotoxic aldehydes.
