Adenylate kinase 5 (AK5) is a cytosolic enzyme belonging to the adenylate kinase family, which plays a crucial role in cellular energy homeostasis by catalyzing the reversible transfer of phosphate groups among adenine nucleotides, thereby regulating the adenine nucleotide composition within cells. Structurally, AK5 is a single polypeptide chain of 586 amino acids with a molecular mass of approximately 65.9 kDa, and it contains a glycine-rich region for nucleoside triphosphate binding as well as a distinct site for nucleoside monophosphate binding. AK5 is expressed predominantly in the brain, where it is involved in energy regulation and cellular differentiation. In terms of disease relevance, altered expression and methylation of AK5 have been linked to various cancers, including breast, gastric, colorectal, and prostate cancers, as well as neurodegenerative disorders such as Alzheimer’s and Parkinson’s disease, autoimmune encephalitis, diabetes, and celiac disease.
